The IsoBind Blood RNA Kit gives clinical and molecular laboratories a manual silica spin-column route to total cellular RNA from 2.5 mL human whole blood collected directly into an EDTA tube.
The sample is treated with 20 mL Gene Vantage Red Blood Cell Lysis Buffer to selectively remove erythrocytes while retaining the nucleated-cell fraction. The recovered cell pellet then undergoes Proteinase K treatment at 56°C before binding conditions are adjusted for capture on the silica membrane. Two wash stages reduce haemoglobin, haeme compounds and other blood-derived contaminants.
Optional on-column DNase treatment may be incorporated when additional genomic DNA removal is required. DNase and its associated treatment reagents are not supplied with the kit. RNA is recovered through two consecutive 50 µL elutions.
The workflow processes 1-24 samples in approximately 45 minutes using a standard laboratory centrifuge. It is intended specifically for 2.5 mL EDTA whole-blood samples and is not intended for plasma, buffy coat or heparin blood.
Purified RNA is suitable for gene expression profiling, biomarker quantification and RT-PCR workflows. The kit is manufactured to international standards and supplied for in vitro diagnostic use.
