Overview

The band you cut from a gel is the material your next experiment depends on, and any agarose or salt that comes with it can stop the ligation, sequencing or PCR that follows.

The IsoBind Gel Extraction Kit recovers your fragment free of the gel, so the next reaction gets clean template.

Key benefits

Keep only the fragment you need
Recovers the single band you cut from the gel, so only your chosen fragment goes into the next step.
Uses equipment you already have
Uses a microcentrifuge, heating block and other standard laboratory equipment, with no dedicated instrument to buy.
Any band you cut
PCR products, restriction fragments and other bands all go through the same protocol.
Concentrated eluate
Elutes in 50 µL, so the recovered fragment stays concentrated for ligation, sequencing or PCR.

How it works

Excise the chosen band and dissolve the agarose. Add binding buffer, load the spin column, then wash, dry and elute the purified fragment.

Workflow

1Excise the selected band2Dissolve the agarose3Prepare the sample for binding4Load the spin column5Wash the membrane6Dry the membrane7Elute the fragment

Before first use

Add ethanol to the wash buffer concentrate and isopropanol to the binding buffer, as described in the manual. Mark each bottle once prepared.

Protect the fragment in your gel

Trim away unnecessary agarose and use blue-light illumination where possible. If using UV light, keep exposure brief to limit damage to the fragment.
