The AmpliMax Small Seed DNA Kit is designed for genomic DNA preparation from tomato, broccoli and onion seeds. The magnetic bead chemistry is designed for high throughput automated processing for breeding, seed-testing and crop-research laboratories.
Effective mechanical disruption and homogenisation are critical for this low-input seed workflow. Because only a small amount of tissue is processed, sample loss during manual grinding and transfer can significantly reduce nucleic acid recovery. We therefore strongly recommend bead milling in robust tubes using appropriately sized 3-5 mm stainless-steel beads. Complete pulverisation increases the surface area exposed to the lysis buffer, supports uniform cell disruption and improves the consistency of nucleic acid release across samples.
A proprietary nucleic acid recovery enhancer supports DNA capture on the magnetic beads. The wash chemistry reduces starch and polysaccharides, while the final wash removes residual detergents and other compounds that could interfere with downstream assays.
The method has been validated on Tianlong Libex and Seegene SEEPrep32. It is also compatible with KingFisher, Revvity chemagic 360, LGC oKtopure and configurable liquid handlers from Hamilton, Beckman Coulter, Tecan and Opentrons. For configurable liquid handlers, the user laboratory is responsible for instrument programming, labware selection and local workflow verification.
