The AmpliMax Plant DNA Kit prepares genomic DNA from supported plant leaf and root samples using magnetic bead chemistry. It can be used for smaller manual batches or transferred to automated systems for routine crop testing and high-throughput breeding programmes.
Effective tissue disruption and homogenisation increase the surface area exposed to the lysis buffer, promoting consistent cell lysis and nucleic acid recovery. Small batches can be ground manually using liquid nitrogen and a mortar and pestle. Higher-throughput laboratories can use a bead mill or tissue homogeniser with 3-5 mm stainless-steel beads in robust tubes or compatible deep-well plates.
A proprietary nucleic acid recovery enhancer supports DNA capture during binding. The wash buffers reduce polyphenols and polysaccharides, while the final wash removes residual detergents and other compounds that could interfere with downstream assays.
The method has been validated on Tianlong Libex and Seegene SEEPrep32. It is also compatible with KingFisher, Revvity chemagic 360, Bioer, Zhengkebio, BioPerfectus, AdNAP, Snibe, and configurable liquid handlers from Hamilton, Beckman Coulter, Tecan, LGC oKtopure and Opentrons. . For configurable liquid handlers, the user laboratory is responsible for instrument programming, labware selection and local workflow verification.
