The IsoBind Maxi Plant DNA Kit provides breeding and agrigenomics laboratories with a high-throughput silica spin-plate workflow for genomic DNA purification from dry plant leaf tissue in 96-well batches.
Ten to 20 mg dry tissue is mechanically disrupted before lysis in 600 µL Nova C-05 with 10 µL Proteinase K at 60°C for 30 minutes. Chloroform clarification separates the aqueous phase, after which Bind C is added at 0.6 times the recovered lysate volume and 10 µL Booster L supports silica binding.
The prepared lysate is loaded onto a 96-well silica spin plate. One 300 µL Wash Nova-Y stage and two 600 µL Wash Ultimus stages reduce polysaccharides, polyphenols and other plant-derived contaminants, followed by a dry spin. DNA is eluted in 50 to 100 µL preheated Elution Buffer, allowing the laboratory to select the volume required for the downstream assay.
The workflow uses a compatible plate centrifuge and does not use magnetic beads. Purified DNA is suitable for high-throughput plant genotyping, GMO testing, variety registration and next generation sequencing.
